Abstract
Nors abscizo rūgštis (ABR) yra gerai žinomas augalų fitohormonas, tačiau trūksta duomenų apie ABR poveikį medžių daigų ir ūglių eksplantų vystymuisi in vitro. Frankia alni poveikis taip pat žinomas, tačiau nenagrinėtas simbiozės efektyvumas esant skirtingoms ABR koncentracijoms. Šio darbo tikslas – patikrinti, ar esant skirtingoms egzogeninėms ABR koncentracijoms in vitro galima valdyti juodalksnio (Alnus glutinosa L.) morfogenezę skirtingose augalo dalyse, tokiose kaip hipokotiliai, epikotiliai, pirminės ir antrinės šaknys. Juodalksnis, kaip tiriamasis objektas pasirinktas, nes gali augti nederlingame ir drėgname dirvožemyje. Augdamas tokiose sąlygose juodalksnis palaipsniui gerina dirvos kokybę, taip suteikdamas sąlygas naujų rūšių augimui. Juodalksnis gali būti pasitelkiamas, kaip priemonė kovoje su dirvos erozija ir klimato kaita. Tyrimas atliekamas trimis etapais. Pirmajame etape juodalksnio sėklos atrenkamos ir dezinfekuojamos. Tada sėklos perkeliamos ant mitybinės WPM terpės mėgintuvėliuose su skirtingomis ABR koncentracijomis: 0 (kontrolė), 1 arba 3 µmol/l. Antrajame etape augalai perkeliami iš mėgintuvėlių į stiklainėlius su terpe be abscizo rūgšties. Prieš perkeliant juodalksnio sėjinukus, pašalinamos jų šaknys ir paliekami 1 cm ilgio ūgliai. Trečiame etape augalai, auginti ant terpės su skirtingomis ABR koncentracijomis, perkeliami į Petri lėkšteles su WPM terpe ir Frankia alni bakterijomis ir lėkšteles be bakterijų. Po 2 savaičių vertinami juodalksnio sėjinukų parametrai. Atlikto tyrimo metu, kai juodalksnio sėklos daiginamos mėgintuvėliuose nustatytas reikšmingas hipokotilių ilgio sumažėjimas ant terpės su 3 µmol/l ABR koncentracija. Taip pat, nustatytas reikšmingas pirminių ir antrinių šaknų ilgių padidėjimas juodalksnio sėklas daiginant ant terpės su 1 µmol/l ir 3 µmol/l ABR koncentracijomis, lyginant su kontroline grupe. Pastebėtas ir išliekamas stimuliuojantis poveikis juodalksnio ūglių eksplantų hipokotilių, pirminių ir antrinių šaknų ilgiui, kai sėjinukai daiginti ant terpės su 3 µmol/l ABR koncentracija, lyginant su kontroline grupe. Tiriant ABR poveikį juodalksniui auginamam su Frankia alni bakterijomis, nustatytas reikšmingas stimuliuojantis poveikis augalo antrinių šaknų ilgiui, kai sėjinukai buvo auginti ant terpės su 1 µmol/l ABR koncentracija.
Althoug abscisic acid (ABA) is a well known phytohormone, there is not enough data about ABA impact on tree sprouts and sprouts explants growth in vitro. Frankia alni effects is known too, but symbiosis efficiency with different fitohormones, such as ABA, concentrations is not defined. This research task is to check if it is possible to control black alder’s (Alnus glutinosa L.) morphogenetic processes in different plant parts, such as hypocotyls, epicotyls, primary and secondary roots, using different abscisic acid concentrations in vitro.
Black alder as exploratory object is selected because it can grow in barren and wet soil. Growing in soil like this black alder gradually improves soil quality, making it possible for a new species to grow in it. Also, black alder can be as a measure to fight with soil erosion and climate change.
Whole research is done in three steps. At the first step we select and disinfect black alder’s seeds. Then seeds are taken to test tubes with WPM, which in its composition respectively has no ABA, 1 µmol/l and 3 µmol/l of ABA. In second step we transfer plants from the test tubes to the glass jars with WPM without ABA. Before transfering black alder’s seedlings we eliminate its roots and keep just 1cm length stems. In the third step plants grown with different ABA concentrations were tranfered to Petri dishes with Frankia alni bacteria and without it.
Study has shown significant hypocotyls length decrease of black alder which were germinated in test tubes with WPM and 3 µmol/l of ABA. Also, significant increase of primary and secondary roots length had been measured on black alder which were germinated on medium with 1 µmol/l and 3 µmol/l ABA concentrations in test tubes, comparing with the control group. Remaining stimulating effect on black alder sprouts explants hypocotils, primary and secondary roots length had been noticed, when plants had been grown on medium with 3 µmol/l ABA concentration.
