Hipochloritinės rūgšties įtaka pelės hepatomos MH–22A ląstelėms in vitro ir jų elektroporacijos tyrimams

Elektroporacijos metu ląstelės yra veikiamos aukštosios įtampos elektriniais impulsais, kurių parametrai parenkami atsižvelgiant į ląstelių tipą bei į tai, kokias manipuliacijas norima su jomis atlikti. Veikiant elektros lauku, ląstelės membranoje susidaro poros, kurios leidžia į ląstelę įnešti įvairias molekules, tokias kaip: DNR, vaistai ar fluorescuojantys dažai. Elektrocheminiai procesai yra neatsiejama elektroporacijos dalis, atliekant tyrimus su ląstelėmis. Šie procesai gali sukelti įvairius pokyčius: terpės pH, cheminės terpės sudėties, temperatūros, elektrodų paviršiaus topografijos pakitimus, aktyviųjų deguonies formų (ROS) terpėje susidarymą. Tai gali daryti poveikį ląstelių gyvybingumui bei ląstelių elektroporacijos efektyvumo vertinimo rezultatams. Šiame darbe pristatomi tyrimai, analizuojant vieną iš elektrocheminių procesų – hipochloritinės rūgšties susidarymą bei jos poveikį pelės hepatomos MH–22A ląstelėms in vitro ir jų elektroporacijos tyrimams. Tam buvo atlikta išsami literatūros analizė apie hipochloritinės rūgšties išsiskyrimą terpėje elektrinių impulsų metu. Šios rūgšties koncentracija terpėje didėja proporcingai didinant elektrinio impulso amplitudę ir/ar impulsų skaičių bei trukmę. Darbe, taikant spektrofluorimetrinės analizės bei mikroskopijos metodus, tirtas hipochloritinės rūgšties bei elektrinio impulso neigiamas poveikis pelės hepatomos MH–22A ląstelių gyvybingumui. Visi darbe atlikti tyrimai gali būti naudingi, norint sumažinti elektrocheminių procesų pasekmes, vykstančias ląstelių elektroporacijos metu.
During electroporation cells are exposed to high–voltage electrical pulses of which parameters are selected according to the type of cells and the type of manipulations desired. By acting on an electric field, pores form in the cell membrane which allows various molecules to be inserted into the cells, such as: DNA, drugs, or fluorescent dyes. In cell–based studies electrochemical processes are an integral part of electroporation. These processes can cause various changes: changes in the pH of the medium, chemical composition, temperature, topography of the electrode surface and formation of active oxygen species (ROS) in the media. This may affect the results of cells’ viability and the results of cells’ electroporation effectiveness evaluation. This paper presents research by analyzing one of the electrochemical processes – the formation of hypochlorous acid and its effect on mouse’s hepatoma MH–22A cells in vitro and their electroporation studies. In order to reach this, a detailed analysis of the literature on the release of hypochlorous acid in the medium during electrical impulses was performed. The increase of hypochlorous acid in the media is proportional to the amplitude of the electrical impulse and/or the number and duration of the impulses. The negative effects of hypochlorous acid and electrical impulse on the viability of mouse’s hepatoma MH–22A cells were studied using spectrofluorimetric analysis and microscopy methods. All the studies performed in the work can be useful to reduce the consequences of electrochemical processes that occur during cell electroporation.
