Dermacentor reticulatus rūšies erkių filogenetinė analizė pagal ITS2 ribosominį DNR fragmentą

Tyrimo problema: Per kelis pastaruosius dešimtmečius yra pastebimas spartus Dermacentor reticulatus rūšies plitimas Europoje. Šios rūšies erkės yra pagrindinis vektorius, Europoje, pernešantis Babesia canis rūšies patogenus, kurie sukelia šuninę babeziozę, kuri kelia didelę grėsmę naminiams gyvūnams. Taip pat trūksta duomenų apie Dermacentor reticulatus genetinę medžiagą. Tyrimo klausimai: Apžvelgti Dermacentor reticulatus paplitimą ir pernešamus patogenus. Atlikti D. reticulatus rūšies erkių ITS2 ribosominio DNR fragmento pagausinimą PGR metodu. Atlikti kitų mokslinių grupių rezultatų paiešką pasinaudojant BLAST algoritmu. Remiantis sekoskaitos duomenimis ir sekomis iš duomenų bazės, atlikti Dermacentor reticulatus filogenetinę analizę pagal ITS2 ribosominį DNR fragmentą. Tyrimo objektas: Dermacentor reticulatus, kitaip žinoma, kaip pievinė erkė. Tyrimo tikslas: Atlikti Dermacentor reticulatus erkių rūšies filogenetinę analizę pagal ITS2 ribosominį DNR fragmentą. Tyrimo metodai: PGR metodu buvo pagausintas D. reticulatus ITS2 ribosominis DNR fragmentas. Atlikta kitų mokslinių grupių rezultatų paieška. Įvykdyta filogenetinė analizė, remiantis sekoskaitos duomenimis ir sekomis iš duomenų bazės. Tyrimo rezultatai / išvados: D. reticulatus geografinis paplitimas yra dalijamas į du makroregionus: vakarų – paplitę šiaurinėje Ispanijos dalyje, Prancūzijoje, Belgijoje, Vengrijoje, Čekijoje, Slovakijoje ir Vokietijoje, o rytų regionas apima – Lenkiją, Baltarusiją, Latviją, Lietuvą ir Rusiją. ITS2 ribosominis DNR fragmentas pagausintas PGR metodu, gautos 34 sekos. GenBank duomenų bazėje buvo aptiktos tik 12 sekų, kurios taip pat buvo įtrauktos į analizę. Analizėje atsiskyrė dvi grupės: pirmoji grupė – individai aptikti Slovakijoje, Dž. Britanijoje, Vokietijoje, Vakarų ir Rytų Lenkijoje; antroji grupė (sekos iš duomenų bazės) – individai aptikti Vokietijoje, Rytų ir Vakarų Lenkijoje ir Ukrainoje. Taip pat atsiskyrė vienas unikalus haplotipas iš duomenų bazės (Vokietija).
Research problem: During the last few decades rapid spread of Dermacentor reticulatus species was observed in Europe. D. reticulatus is the main vector for Babesia canis, which is the cause for babesiosis in pets. In-addition there is lack of information on Dermacentor reticulatus genetic material. Research questions: Examine prevalence and transmittable pathogens of Dermacentor reticulatus. Carry out Dermacentor reticulatus tick species amplification of ITS2 ribosomal DNA fragment using PCR method. Perform other scientist groups results search using BLAST algorithm. Using sequencing data and sequences from database, carry out Dermacentor reticulatus phylogenetic analysis of ITS2 ribosomal DNA fragment. Research object: Dermacentor reticulatus, also known as meadow tick. Research aim: Carry out Dermacentor reticulatus tick species phylogenetic analysis according to ITS2 ribosomal DNA fragment. Research methods: D. reticulatus ITS2 ribosomal fragment was amplified by using PCR method. Other scientific groups results overview was carried out. Phylogenetic analysis was done according to sequencing data and sequences from database. Research results / conclusions: D. reticulatus geographical distribution is split into two macroregions: west – prevalent in northern Spain, France, Belgium, Hungary, Czech Republic, Slovakia, and Germany; east region – Poland, Belarus, Latvia, Lithuania and Russia. ITS2 ribosomal DNA fragment was amplified using PCR and 34 sequences were obtained. Only 12 sequences were found in GenBank database which were used in our analysis. The results of analysis were secluded into two groups: first one – individuals found in Slovakia, Great Britain, Germany, western and eastern Poland; second group (sequences from database) – individuals found in Germany, eastern and western Poland and Ukraine. There was a single unique haplotype from database (Germany).
