Simbiotinės augalų bakterijos Paenibacillus sp. įtakos europinio maumedžio (Larix decidua Mill.) morfogenezei tyrimas viršūninių pumpurų in vitro kultūrose
Baigiamojo darbo tikslas - ištirti maitinamojoje terpėje esančios simbiotinės augalų bakterijos Paenibacillus sp. poveikį europinio maumedžio viršūninių pumpurų eksplantų morfologiniams parametrams. Europinis maumedis (Larix decidua Mill.) dėl savo vertingų savybių yra perspektyvus mikrodauginimo in vitro objektas, tad šiuo darbu buvo siekiama patikrinti, ar maumedžio vystymasi galima sustiprinti simbiotinių bakterijų pagalba. Tiriamoji bakterija buvo iš hibridinės drebulės (Populus tremuloides × P. tremula) audinių kultūros išskirta Paenibacillus sp. Iki šiol galimybės sustiprinti europinio maumedžio vystymąsi pasitelkiant Paenibacillus genties bakterijas nebuvo tirtos. In vitro sistemoje buvo tiriamas Paenibacillus genties bakterijos poveikis viršūninių europinio maumedžio eksplantų vystymuisi, priklausomai nuo bakterijos auginimo sąlygų bei auginimo trukmės prieš eksperimentą. Gauti rezultatai rodo, kad bakterija europinio maumedžio eksplantų augimą veikia teigiamai, kai prieš jų sąveiką in vitro kultūroje, bakterija yra auginama jai nepritaikytomis sąlygomis ir jai sukeliamas priverstinis ramybės būsenos palaikymas. Europinio maumedžio eksplantus paveikus su tokiomis sąlygomis auginta bakterija registruojami 137 proc. ilgesni ūgliai (p < 0,001) bei 135 proc. ilgesni spygliai (p < 0,001). Atsižvelgiant į tai, bakterijos gebėjimas išskirti tam tikras augalų augimą skatinančias chemines medžiagas gali būti siūlomas kaip perspektyvus diskusijų ir tolesnių tyrimų objektas.
The aim of the master thesis was to determine the influence of the symbiotic bacteria Paenibacillus sp. on the viability of european larch (Larix decidua Mill.) buds. Due to its valuable properties European larch (Larix decidua Mill.) is a promising object for in vitro micropropagation, so the aim of this work was to test whether the development of larch can be enhanced by symbiotic bacteria. The tested bacteria was isolated first from the hybrid aspen (Populus tremuloides × P. tremula) tissue culture and identified as Paenibacillus sp. The potential for enhancing the development of European larch by symbiotic Paenibacillus bacteria has not been investigated. The in vitro system was used to study the effect of Paenibacillus on the development of European larch explants, depending on the growth conditions and growth duration of the bacterium before the experiment. The results kad shown that with the help of the Paenibacillus sp. bacteria it is possible to successfully grow shoots from vegetative buds of European larch isolated in vitro. It has been found that bacteria has a positive effect on the growth of European larch explants when bacteria is grown under conditions unsuitable conditions and is forced to dormancy, before their interaction in one in vitro culture. When European larch explants were exposed to bacteria grown under these conditions, 137 pct. longer shoots (P < 0.001) and 135 pct. longer needles (P < 0.001) were recorded. In this context, the bacteria’s ability to release certain plant growth-promoting chemical might be suggested as a promising object further research.
