Research was carried out in the Laboratory of Agrobiotechnology of Institute of Biology and Plant Biotechnology in the Faculty of Agronomy at the Vytautas Magnus University in 2018–2019. In this research the opium poppy (Papaver somniferum) genotypes ‘Hen and chickens’ and ‘Danish flag’ explants – leaf, cotyledons and hypocotyls – were cultivated in nutrition medium supplemented with 0.2; 0.5; 1.0 mg l-1 α-naphthylacetic acid (NAA) and 0.1 mg l-1 kinetin (KIN) combinations. The results showed that callus induction in genotype ‘Danish flag’ were about 1.4 times more intensive than in genotype ‘Hen and chickens’. The isolated leaves explants of genotype ‘Hen and chickens’ formed callus in higher frequency than cotyledons and hypocotyls. The isolated cotyledons explants of genotype ‘Danish flag’ were higher in cells dedifferentiation potency than leaves and hypocotyls. For callus induction of genotype ‘Hen and chickens’ recommended to cultivate leaves explants in nutrition medium supplemented with 1.0 mg l-1 NAA and 0.1 mg l-1 KIN, while genotype ‘Danish flag’ – hypocotyls explants in nutrition medium supplemented with 0.5 mg l-1 NAA and 0.1 mg l-1 KIN.
