The main goal of the research was to evaluate the influence of various plant hormones on callus formation from leaf explants in Zantedeschia genus. The research was carried out in 2015–2016 at ASU Biology and plant biotechnology institute with Zantedeschia aethiopica cultivars. Leaf explants were planted on Murashige and Skoog (MS) medium supplemented with different plant growth regulator concentrations and combinations. Firstly, two different cytokinins were tested, BAP and KIN. It was found that 2.0 BAP mg l-1 was most efficient, this optimal concentration developed an average of 58.4 % of callus formation. Other cytokinin concentrations and types were not that effective. Second best rate was obtained in medium with 1.0 mg l-1 KIN, where 44.7 % of all leaf explants have formed callus. Other cytokinin concentrations showed quick fall of callus formation rate and worst results were obtained in medium with 1.0 mg l-1 BAP, where only 11,1 % of all leaf explants have formed callus. In later stage of research, additional concentrations of auxins were added to the MS medium with 2.0 BAP mg l-1, which in some cases enhanced callus formation. Callus formation was most efficient with 0.5 IAR+2.0 BAP mg l-1, where up to 81,6 % of all explants formed a callus. Callus formation was enhanced in medium supplemented with 0.25 ISR+2.0 BAP mg l-1, rate got up to 61.6 % and in medium with 0.5 mg l-1 ISR+2.0 BAP mg l-1, where rate was respectively 70.2 %. It is also important to note, that explants, which were planted on MS medium without plant growth regulators, could not form callus. This means, that growth regulators are crucial in dediferentation of Zantedeschia leaf explants. This research was carried out under controlled conditions an al explants were sterilised.
