This work presents the influence of different plant growth regulators on Miscanthus x giganteus Greef et Deuter callus induction in vitro. Research was carried out in 2015–2016 in the JRC laboratory of Agrobiotechnology, Institute of Biology and plant biotechnology of Aleksandras Stulginskis university. Method of the work – isolated root explants were sterilized and grown on MS nutrient medium without and with different quantities of selected growth regulators – IBA, 2.4-D and 2iP. MS medium was supplemented with 10 g l-1 sucrose and 8 g l-1 Difco Bacto-agar. The media pH was 5.8. Isolated root explants were grown in vitro at 22 ± 2 °C temperature, 50 μmol m-2s-1 light intensity of 16/8h (day/night) photoperiod. After 30 days percentage of callus formation frequency was calculated. The results of the work – Miscanthus x giganteus roots tissues can form callus without growth regulators with a low frequency. Callus induction were determined in all selected media, but callus formation process depended on the intensity of auxins (IBA or 2.4-D) and cytokinin (2iP) in media. The best combinations of growth regulators were 5.0 mg l-1 IBA + 0.1 mg l-1 2iP and 5.0 mg l-1 2.4-D + 0.1 mg l-1 2iP. Under these conditions, the callus induction percentages were 58.5 % and 62.4 %.
