Investigations were carred out during 2012–2013 in the Laboratory of Agrobiotechnology and Institute of Biology and Plant Biotechnology in the Faculty of Agronomy at the Aleksandras Stulginskis University. The aim of work: to evaluate the growth regulator combinations on St. John's wort shoot formation in vitro. The results of this study shows that in medium without growth regulators St. John's wort isolated stem segments formed shoots with low frequency. Investigated combinations of cytokinin and auxins in medium substantially promoted St. John's wort shoot formation in vitro. The medium supplement by 0.1 mg l-1 BAP + 4.5 mg l-1 NAA resulted in highest shoot formation frequency (100 %) and the highest number of shoots per explant (47.86 unit).
