Organogenesis induction in leaf explants of Sedum L

Propagation of ornamental plants throughout in vitro techniques is important for a wide range of application. Plant regeneration was studied in leaves culture of Sedum rupestre ‘Angeline’, Sedum reflexum ‘Cristatum’, Sedum fosterianum ‘Elegans’, Sedum maximum ‘Autumn Joy’. The effect of different levels of 6-benzylaminopurine (BAP) and 1-naphthylacetic acid (NAA) on plant regeneration was evaluated. Isolated explants were growing on Murashige and Skoog (MS) nutrition medium with different content of growth regulators. Variable organogenic responses were expressed by all of the cultivars tested on the different culture medium. Significant differences were observed in the mean level of shoot formation on responsive culture media. The maximum number of shoots per explant from cultivars ‘Elegans’ and ‘Angeline’ was obtained in a medium supplemented with 0.75 mg l-1 BAP. The use of combinations 3.0 mg l-1 BAP + 0.5 mg l-1 NAA and 4.0 mg l-1 BAP + 0.5 mg l-1 NAA significantly increased number of shoots per explant from leafs segments of ‘Autumn Joy’ while the 3.0 mg l-1 BAP induced the highest number of shoots from explants of ‘Cristatum’. The highest shoots number per explant of variety ‘Angeline’ has been establishing in medium with 0.75 mg l-1 BAP and 3.0 mg l-1 BAP. Medium supplemented with 3.0 mg l-1 BAP resulted in highest shoots number per explant of ‘Cristatum’. Explants of cultivar ‘Autumn Joy’ highest shoots number per explant formed in medium with 3.0 mg l-1 BAP + 0.5 mg l-1 NAA and 4.0 mg l-1 BAP + 0.5 mg l-1 NAA. Adventitious shoots were transferred to MS culture medium supplemented with 0.1 mg l-1 NAA for rhizogenesis induction. Well rooted planlets were acclimatized and subsequently established in soil.
