Theoretical background and experimental measurements of estimation probability electroporation of needed amount of the cells in suspension as a function of electric field parameters are presented. Three steps in process of high electric field pulse generators-electroporators construction, manufacturing and testing carried out in our laboratory are discussed. Aiming to measure theoretically predicted probability of electroporation cells in vitro the erythrocytes and hepatoma MH22 line cells were used. The electroporation efficiency was estimated by two different methods: (i) measuring efflux of intracellular K+ by using mini selective and reference electrodes (ii) counting the cells killed by anticancer drug bleomycin. It was shown that on the basis measured electroporation probability versus electric field parameters, the optimal set of pulse strengthduration combinations allowing transfer of chosen small molecules into defined amount of cells could be predicted and the size of electropores could be estimated.
