Kalceino fluorescencijos slopinimas chromo (VI) jonais elektroporuotose ir neelektroporuotose pelės hepatomos MH - 22A ląstelėse

Elektroporacija – tai porų formavimasis ląstelių membranose dėl išorinio elektrinio lauko padidinto membraninio potencialo. Ląstelėms elektroporuoti naudojami aukštosios įtampos impulsai, kurių metu ląstelių suspensija teka didelė elektros srovė ir vyksta elektrocheminiai procesai. Dėl šalutinių reakcijų gali pastebimai keistis terpės pH, temperatūra, cheminė sudėtis bei elektrodų paviršius. Iš elektrodų išsiskyrusių metalų jonai patekę į terpę yra toksiški ir kartu su tikslinėmis molekulėmis (DNR, vaistai ar kt.) patekę į ląstelės vidų gali neigiamai paveikti ląstelių gyvybingumą. Elektrodai išskiria jonus, kurie įeina į jų sudėtį pvz. nerūdijančio plieno elektrodai išskiria Fe2+/Fe3+, Cr, Ni, Mn jonus. Šiame bakalauro darbe buvo atliekami eksperimentai, siekiant nustatyti chromo (IV) jonų, išsiskiriančių iš elektrodų elektromanipuliacijų su ląstelėmis metu, galimą įtaką kalceino fluorescencijos intensyvumui pelės hepatomos MH – 22A ląstelėse. Pirma buvo nustatyta, kad chromo (VI) jonai slopina kalceino fluorescenciją PBS terpėje. Toliau buvo tiriama aukštosios įtampos impulsų įtaka su kalceino – AM dažu pripildytų pelės hepatomos MH – 22A ląstelių fluorescencijai. Rezultatai parodė, kad pritaikius 2 ms trukmės 1 kV/cm vienetinį stačiakampį impulsą, ląstelių fluorescencijos intensyvumas sumažėjo iki 63 %, o esant 1,5 kV/cm elektrinio lauko intensyvumui – iki 39 %. Taip pat darbe ištirta, kad chromo (IV) jonai slopina elektroporuotų ir intaktinių pelės hepatomos MH – 22A ląstelių fluorescenciją.
Electroporation is the process where pores are forming in cell membranes due to increased membrane potential of an external electric field. High - voltage pulses are used to electroporate cells, during which a large electric current flows through the cell suspension. Side reactions can cause significant changes in the pH, temperature, chemical composition of the medium and electrode surface. Metal ions released from the electrodes are toxic, and together with the target molecules (DNA, medicine, etc.), they can negatively affect the viability of the cells. Electrodes emit ions that they consist of, e.g., stainless steel electrodes emit Fe2 + / Fe3 +, Cr, Ni, Mn ions. In this study, experiments were performed to evaluate the possible influence of chromium (VI) ions released from the electrodes during electromanipulation of cells with the intensity of calcein – AM fluorescence in mouse hepatoma MH – 22A cells. It was first determined that chromium (VI) ions quench the intensity of calcein fluorescence in PBS medium. Then the influence of high voltage pulses on the fluorescence of mouse hepatoma MH - 22A cells was investigated. The results showed that after applying a 2 ms 1 kV / cm single rectangular pulse, the fluorescence intensity of the cells decreased to 63 % and after 1.5 kV / cm electric field intensity it decreased till 39 %. Finally, the effect of chromium (VI) ions on the fluorescence of electroporated and intact mouse hepatoma MH - 22A cells was determined.
