Abstract
Magistratūros studijų baigiamajame darbe pateikiama 2017 – 2018 m. Vytauto Didžiojo universiteto Žemės ūkio akademijos Agronomijos fakulteto Biologijos ir augalų biotechnologijos instituto Agrobiotechnologijos laboratorijoje tirtų maitinamosios terpės komponentų poveikio žieminio rapso augimui kintančios temperatūros sąlygomis in vitro, tyrimo rezultatai.
Darbo objektas – žieminis rapsas (Brassica napus var. oleifera L.)
Darbo metodai: tirtų žieminių rapsų veislių ‘Visby’, ‘Mercedes’, ‘Kuga’ ir ‘Anderson’ augalai auginti skirtingose maitinamosiose terpėse: MS (be augimo reguliatorių); MS+ 3,0 mg l–1 BAP; MS+ 3,0 mg l–1 BAP + 60,0 g l–1 sacharozės; MS+ 0,8 mg l–1 B1+ 1,0 mg l–1 B6+ 1,0 mg l–1 PP ; MS+ 0,8 mg l–1 B1+ 1,0 mg l–1 B6+ 1,0 mg l–1 PP +60,0 g l–1 sacharozės; MS+ 60,0 g l–1 sacharozės. Vertintas žieminio rapso –įšgyvenamumas terpėse po grūdinimo (proc.), ūglių atsparumas šalčio poveikiui (proc.) ir žieminio rapso genotipų antžeminės dalies ilgis (cm) in vitro.
Darbo rezultatai. Tirtų žieminių rapsų augalų atsparumas šalčiui in vitro priklauso nuo genotipo, vitaminų priedo maitinamojoje terpėje ir padidinto sacharozės kiekio, veislės žiemkentiškumo ir šaldymo temperatūros. Iš tirtų žieminių rapsų genotipų didžiausias išgyvenusių augalų procentas gautas grūdinant visose tirtose temperatūrose (4, 6, 8 °C) veislių ‘Mercedes’ ir ‘Anderson’ augalus. Tyrimais nustatyta, kad tiriant temperatūrų (–2, –6, –10 °C) pokyčius, žiemkentiškiausia veislė – ‘Mercedes’. Didžiausias nepažeistų augalų procentas gautas augalus auginant terpėje su vitaminų 0,8 mg l–1 B1+ 1,0 mg l–1 B6+ 1,0 mg l–1 PP priedu. Nustatyta, kad didžiausią (42,5 cm) antžeminę dalį užaugino veislės ‘Mercedes’ augalai auginti terpėje su vitaminais 0,8 mg l–1 B1+ 1,0 mg l–1 B6+ 1,0 mg l–1 PP ir padidintu 60,0 g l–1 sacharozės kiekiu.

The effect of nutrient medium composition on the growth of winter rapeseed under changing temperature conditions. Research was investigated in the Laboratory of Agrobiotechnology of Institute of Biology and Plant Biotechnology in the Faculty of Agronomy at the Aleksandras Stulginskis University (Vytautas Magnus University Agricultural academy) in 2017 – 2018.
Object of the work – winter rape (Brassica napus var. oleifera L.)
Method of the work – The plants of the winter rapeseed varieties Visby’, ‘Mercedes’, ‘Kuga’ and ‘Anderson’ were grown in different nutrient media. Shoots to grow MS (without growth regulator); MS + 3.0 mg l-1 BAP; MS + 3.0 mg l-1 BAP + 60.0 g l-1 sucrose; MS + 0.8 mg l-1 B1, 1.0 mg l-1 B6, 1.0 mg l-1 PP; MS + 0.8 mg l-1 B1, 1.0 mg l-1 B6, 1.0 mg l-1 PP + 60.0 g l-1 sucrose; MS + 60.0 g l-1 sucrose. Estimated winter rape survival in different nutrient media after hardening (%), shoots resistance to cold temperature (%) and winter rape genotype overhead (cm.).
Results of the work – The in vitro resistance of the winter rape shoots tested depends on the genotype, the exogenous vitamins additive in the nutrient medium and the increased sucrose content, the wintering qualities of the variety and the freezing temperature. The highest percentage of surviving shoots in the winter rape genotypes tested was obtained by tempering the 'Mercedes' and 'Anderson' shoots at all tested temperatures (4, 6, 8 ° C). Studies have shown the variation in temperatures (–2, –6, –10 ° C) is the most wintered variety, 'Mercedes'. The highest percentage of intact shoots was obtained by growing shoots in the medium containing vitamins 0.8 mg l-1 B1 + 1.0 mg l-1 B6 + 1.0 mg l-1 PP. It was found that the largest (42.5 cm) ground part was grown by the cultivars of 'Mercedes' cultivated with vitamins 0.8 mg l-1 B1 + 1.0 mg l-1 B6 + 1.0 mg l-1 PP and increased by 60, 0 mg l -1 for sucrose.
