Abstract
Amplekso raudonasis – tai fluorescencinis indikatorius, skirtas vandenilio peroksidui nustatyti. Amplekso raudonasis, esant vandenilio peroksidui ir dalyvaujant fermentui krieno peroksidazei, oksiduojamas į rezorufiną, fluorescencinę molekulę. Jos fluorescencija nusako vandenilio peroksido buvimą mėginyje. Bet mėginyje esantis vandenilio peroksidas gali lengvai reaguoti ir su jo aplinkoje esančiais metalų jonais, tokiais kaip Fe3+, Ni2+ ar Cr6+, dalyvaujant Fentono reakcijose, ir taip sumažinti vandenilio peroksido kiekį mėginyje. Tyrimo metu nustatyta, kad iš trijų tirtų metalų jonų (Fe3+, Ni2+ ir Cr6+), fluorescenciją labiausiai mažina geležies (III) jonai, o mažiausią įtaką jai daro – nikelio (II) jonai.
Geležies jonams reaguojant su vandenilio peroksidu, susidaro hidroksilo radikalas, kuris sukelia ląstelėje pažaidas, galinčias lemti ląstelių mirtį. Ištyrus H2O2 ir Fe3+ poveikį kininio žiurkėnuko (lot. Cricetulus griseus) kiaušidžių (angl. Chinese hamster ovary (CHO)) ląstelėms, nustatyta, kad jie šias ląsteles veikia toksiškai.
Geležies jonams reaguojant su vandenilio peroksidu, susidaro hidroksilo radikalas, kuris sukelia ląstelėje pažaidas, galinčias lemti ląstelių mirtį. Ištyrus H2O2 ir Fe3+ poveikį kininio žiurkėnuko (lot. Cricetulus griseus) kiaušidžių (angl. Chinese hamster ovary (CHO)) ląstelėms, nustatyta, kad jie šias ląsteles veikia toksiškai.
Amplex Red is a fluorescence indicator, which is used for the detection of hydrogen peroxide. Amplex Red is oxidised by hydrogen peroxidase in the presence of enzyme horseradish peroxidase originates fluorescent molecule resorufin. The fluorescence of resorufin informs about hydrogen peroxide in the sample. But if metal ions, such as Fe3+, Ni2+ or Cr6+, are present in the sample they react with hydrogen peroxide while forming Fenton reactions thus lowering levels of hydrogen peroxide in the sample. In this work it was established that, out of three metal ions tested here (Fe3+, Ni2+ ir Cr6+), iron (III) ions demonstrated the highest influences on the fluorescence of Amplex Red fluorescence. On the contrary, nickel (II) was the one, which showed the least influence on Amplex Red fluorescence.
