Nuolat auganti nukleozidų paklausa skatina ieškoti įvairių jų sintezės būdų. Šiuo metu nukleozidai yra arba gaunami iš natūralių šaltinių, arba sintetinami cheminiu būdu. Vienas iš labiausiai tyrinėjamų būdų – fermentinė nukleozidų sintezė, kuri, lyginant su chemine sinteze, pranaši tuo, jog reakcijos vyksta selektyviai, susidaro mažiau organinių atliekų.
Šio darbo tyrimo objektas – fermentinė pakopinė reakcija, kurios metu iš deoksiribozės ir azotinės bazės sintetinami deoksiribonukleozidai. Tiriamą pakopinę reakciją vykdo trijų tipų fermentai: ribokinazė, fosfopentomutazė ir nukleozidų fosforilazė. Darbe nagrinėjami fermentai iš bakterijų ir termofilinių mikroorganizmų. Termofiliniai fermentai pasirinkti dėl jų gebėjimo vykdyti reakcijas esant aukštesnėms temperatūroms. Reakcijos esant aukštesnėms temperatūroms naudingos dėl mažesnės mikrobinio užterštumo rizikos, taip pat didėja reagentų tirpumas bei mažėja terpės klampa.
Darbo metu sukonstruoti DNR plazmidiniai vektoriai su pasirinktus fermentus koduojančiais genais, vykdyta tikslinių baltymų raiška. Išgryninti fermentai charakterizuoti – nustatytos koncentracijos bei įvertinti jų aktyvumai. Tiriamieji fermentai įvertinti pakopinėje deoksiribonukleozidų sintezės reakcijoje. Nustatyti reakcijos produktų susidarymo efektyvumai tiek naudojant bakterijų fermentus, tiek termofilinių organizmų fermentus. Įvertinti termofilinių fermentų vykdomų reakcijų efektyvumai, esant skirtingoms temperatūroms. Taip pat nustatyta reakcijos produktų kitimo priklausomybė nuo vykdomos reakcijos trukmės.

The ever-increasing demand for nucleosides promotes the search for a variety of synthesis pathways. Nucleosides are currently either derived from natural sources or chemically synthesized. Enzymatic nucleoside synthesis is under extensive investigation which suggests opportunity to perform reactions more selectively and with less organic waste compared with chemical synthesis.
The subject of this research is coupled enzymatic reaction in which deoxyribonucleosides are synthesized from deoxyribose and heterocyclic base. Three types of enzymes are involved in this reaction: ribokinase, phosphopentomutase and nucleoside phosphorylase. In this thesis the enzymes found in bacteria and thermophilic microorganisms are investigated. Thermophilic enzymes are objects of interest due to their ability to perform reactions at high temperatures. Reactions at higher temperatures are beneficial for lower risk of microbial contamination, as well as the higher solubility of reagents and the reduced viscosity of the medium.
DNA plasmid vectors with genes encoding selected enzymes were constructed. Expression systems for these vectors where developed. Enzymes were purified and then characterized – concentrations were determined and activities were evaluated. Enzymes of interest were used in multienzymatic deoxyribonucleoside synthesis reaction. Efficiency of the reaction to produce desired products was determined both by using bacteria enzymes and thermophilic ones. Efficiency of the reaction using thermophilic enzymes at different temperatures was evaluated. Product formation along reaction time was also determined.
