Abstract
Šiame darbe tirtas elektroporacijos poveikis E.coli O157:H7 bakterijų linijos inaktyvacijai pieno terpėje. Buvo nustatyta, kad pritaikius 2.4 kV/cm elektros įtampą, kai impulso trukmė yra 25µs, o impulsų skaičius 20, poveikis bakterijų inaktyvacijai buvo stipriausias. Bakterijų gyvybingumas lyginant su kontrole, inkubuojant bakterijas 37°C temperatūroje 24 valandas, siekė tik 0.19%.
Tyrimo metu analizuotas ir standartinių pieno pasterizavimo metodų efektyvumas. Nustatyta, kad taikant kombinuotą mikrofiltracijos (0.5 µm porų filtru) ir trumpo išlaikymo aukštos temperatūros (80°C temperatūroje, 20 sekundžių), ilgo išlaikymo žemos temperatūros (65°C temperatūroje, 30 minučių) bei trumpo išlaikymo aukštos temperatūros (80°C temperatūroje, 20 sekundžių) metodus, bakterijų gyvybingumas buvo lygus 0 %. Bakterijų ir pieno suspensiją mikrofiltruojant (0.5 µm porų filtru) gautas 3.25% bakterijų gyvybingumas, lyginant su kontrole. Pasterizuojant (90°C temperatūroje 2 sekundes) bakterijų inaktyvacijai didelės įtakos neturėjo, gyvybingumas išliko santykinai didelis 81.64 %.

In this work, the effect of electroporation on bacterial inactivation of E.coli O157: H7 in the milk medium was investigated. It was found that applying an electrical field strength of 2.4 kV / cm, with a pulse duration of 25 μs and a number of pulses equal to 20, had the strongest effect on bacterial inactivation. The viability of the bacteria compared with the control, after incubation at 37 ° C for 24 hours, was only 0.19%.
In this study the effectiveness of standard milk pasteurisation methods was also analyzed. It was determined that using a combination microfiltration (0.5 μm pore filter) and short-term high temperature (80 ° C, 20 seconds), long-term low temperature (65 ° C, 30 minutes) and short-term high temperature (80 ° C, 20 seconds) pasteurization methods, the viability of the bacteria was 0%. Microfiltration of bacteria and milk using 0.5 μm pore filter yielded to viability of 3.25% compared to control. Pasteurization (at 90 ° C for 2 seconds) did not have any significant effect on bacterial inactivation, viability remained relatively high at 81.64%.
