Abstract
Non-small cell lung cancer (NSCLC) effectiveness is more than 85% of cancers worldwide with an overall survival rate even less than 20%. To date, the complex molecular mechanisms of NSCLC are still unknown, which makes it extremely important to uncover the underlying genes contributed to NSCLC. Therefore, identifying novel biomarkers for diagnosis and prognosis of NSCLC is crucial so that patients can get appropriate treatment as soon as possible. Using GEO (Gene Expression Omnibus database) four datasets (GSE21933, GSE89047, GSE10072, GSE18842) were gathered. DEGs (Differently Expressed Genes) between normal and NSCLC samples were analyzed by utilizing Gene Expression Omnibus 2R (GEO2R) to conduct gene integration and Limma package. Cytoscape, STRING and MCODE were used to make PPI (Protein-Protein Interaction) of the top screened DEGs. Next, Pathway enrichment analysis and functional enrichment for DEGs were analyzed by utilizing OmicShare and FunRich, and finally prognostic value basis and high expressions of acquired genes were carried out by using KM (Kaplan Meier-plotter) and GEPIA (Gene expression profiling interactive analysis) online dataset. Including 428 upregulated and 336 downregulated, a total of 764 DEGs were identified and carried out after gene interactions. PPI network was made, 1784 protein pairs and 165 nodes with >0.4 P-values. In module no. 1, TOP2A (Topoisomerase II Alpha) made a top hub node and gene with high mitotic rich cell cycle pathway along with IL-6 as enriched in amb2 integrin signaling pathway. Five biomarker genes with high degree of connectivity were chosen having worse NSCLC based OS, (CCNB1, CCNA2, TOP2A, KIF20A, UBE2C).
Nesmulkialąstelinis plaučių vėžys (NSPV) sudaro daugiau nei 85% visų plaučių vėžio atvejų pasaulyje, o jo patogenezėje nedaug biologinės informacijos bei bendras išgyvenamumas – mažesnis nei 20%. Iki šiol sudėtingi NSPV molekuliniai mechanizmai vis dar nežinomi, todėl labai svarbu atskleisti pagrindinius genus, prisidėjusius prie NSPV. Taip pat būtina nustatyti diagnozei ir prognozei naujus NSPV biologinius žymenis, kad pacientai galėtų kuo greičiau gauti tinkamą gydymą.Naudojant GEO („Gene Expression Omnibus“ duomenų bazę) buvo surinkti keturi duomenų rinkiniai (GSE21933, GSE89047, GSE10072, GSE18842). DEG (skirtingai išreikšti genai) tarp normalių ir NSPV mėginių, naudojant genų ekspresiją „Omnibus 2R“ („GEO2R“) genų integracijai atlikti ir „Limma“ paketą analizei atlikti. „Cytoscape“, STRING ir MCODE buvo naudojami „PPI“ (Protien-Protien Interaction) gamybai. Toliau buvo atliekama kelio praturtinimo analizė ir funkcinis praturtinimas DEG analizavimui, naudojant „OmicShare“ ir „FunRich“, o prognostinių verčių pagrindas ir didelės įgytų genų išraiškos buvo atliktos naudojant KM („Kaplan Meier-plotter“) ir „GEPIA“ (interaktyvi „Gene expression profiling“ analizė) duomenų rinkinius. Iš viso po genų sąveikos buvo nustatyti 764 DEG, iš kurių (428 padidinti ir 336 sumažinti). Buvo sukurtas PPI tinklas, 165 mazgai ir 1784 baltymų poros su> 0,4 P verte. Modulyje Nr. 1, TOP2A (topoizomerazė II alfa) padarė viršutinį pagrindinį mazgą ir geną, turintį daug mitozės ląstelių ciklo kelią kartu su IL-6, praturtintą amb2 integrino signalizavimo keliu. Buvo pasirinkti penki biomarkerio genai pasižymintys aukštu ryšiu, turintys blogesnę NSPV pagrįstą OS (CCNB1, CCNA2, TOP2A, KIF20A, UBE2C).
