Abstract
Varis – vienas plačiausiai aplinkoje paplitusių sunkiųjų metalų. Herbicidas glifosatas – taip pat viena dažniausiai naudojamų medžiagų piktžolių naikinimui.Dėl neracionalaus jų naudojimo tiek vandens, tiek sausumos ekosistemoms yra daromas neigiamas poveikis. Šio darbo tikslas – ištirti vario (Cu) ir glifosato pavienį bei jų mišinio kompleksinį poveikį netaikinėms sausumos ir vandens augalų rūšims: vasariniams miežiams (Hordeum vulgare L.) ir mažajai plūdenai (Lemna minor L.). Siekiant įvertinti vario ir glifosato diferencijuotą ir kompleksinį jų mišinių poveikį, buvo paruošti 5 skirtingos vario ir glifosato (1μg/l; 5μg/l; 10μg/l; 100 μg/l; 1000μg/l) koncentracijos tirpalai vandens augalams. Kompleksiniam metalo ir herbicido poveikiui mažajai plūdenai (Lemna minor L.) buvo pasirinktos 5 koncentracijos, kurias sudarė vario (Cu) ir glifosato (Gly) mišinys: 0,5μg/l Cu+ 0,5μg/l Gly; 2,5μg/l Cu +2,5 μg/l Gly; 5 μg/l Cu + 5 μg/l Gly; 50 μg/l Cu + 50 μg/l Gly; 500 μg/l Cu + 500 μg/l Gly. Tuo pačiu metu augalai sudedami į kontrolinį tirpalą (Steinberg terpė) be testuojamų medžiagų. Sausumos augalams vassariniams miežiams (Hordeum vulgare L.) buvo tirtos 5 skirtingos vario ir glifosato koncentracijos 1000 kartų stipresnės nei vandens augalams: 0,01mg/kg; 0,1 mg/kg; 1,0 mg/kg; 10,0 mg/kg; 100 mg/kg. Medžiagų mišino koncentracijos sausumos augalams buvo tokios: 0,005 mg/kg Cu+ 0,005 mg/kg Gly; 0,05 mg/kg Cu +0,05 mg/kg Gly; 0,5 mg/kg Cu + 0,5 mg/kg Gly; 5 mg/kg Cu + 5 mg/kg Gly; 50 mg/kg Cu + 50 mg/kg Gly. Kontroliniai augalai buvo laistomi distiliuotu vadeniu. Po 10 parų Lemna minor ir Hordum vulgare ekspozicijos variu ir glifosatu buvo ištirti morfometriniai, fiziologiniai ir biocheminiai augalų rodikliai. Taip pat buvo apskaičiuotos mažųjų plūdenų ir vasarinių miežių EC50 - koncentracijos, sukeliančios 50% atsaką į poveikį kiekvienam teršalui, o mišinio koncentracijos buvo išreikštos pagal toksiškumo vienetų metodą (TU). Įvertinus vario ir glifosato įtaką Lemna minor sausos masės prieaugiui gauta: Cu EC50 = 237,85 μg/l, Gly EC50 = 262,94 μg/l, Cu+Gly = 0,45 TU, o vasariniams miežiams: Cu EC50 = 0,087 mg/kg, Gly EC50 = 5,41 mg/kg, Cu+Gly = 0,64 TU. Taigi, jautresni buvo vandens augalai, tačiau abiem tirtais atvejais augalų sausos masės prieaugį toksiškiau veikė varis - mažesnė jo koncentracija sukelia didesnį slopinimą sausajai biomasei lyginant su tokia pačia glifosato koncentracija. Eksperimento metu nustatyta, jog didžiausią neigiamą poveikį mažųjų plūdenų ir vasarinių miežių augimui, fiziologiniams bei biocheminiams rodikliams darė stipriausios vario ir glifosato koncentracijos (1000μg/l; 100 mg/kg). Medžiagoms veikiant mišinyje, beveik visais tirtais atvejais buvo nustatyta sinergistinė sąveika. Oksidaciniam stresui reikšmingos įtakos turėjo visos tirtos vario ir glifosato koncentracijos. Apibendrinant galima teigti, kad jautresni vario ir glifosato poveikiui buvo vandens augalai (Lemna minor L.).
Copper – is the most common heavy metal in the environment and glyphosate - is the most used herbicide in the world. Glyphosate and copper may pose negative impact on aquatic and terrestrial ecosystems because of unsustainable use of it. The aim of this study was to investigate single and mixture effects of herbicide glyphosate and heavy metal copper to non target terrestrial and aquatic plant species: summer barley (Hordeum vulgare L.) and a common duckweed (Lemna minor L.). In order to investigate effects of copper and glyphosate five different glyphosate and copper (1μg/l; 5μg/l; 10μg/l; 100 μg/l; 1000μg/l) concentrations were prepared of aquatic plants. For complex mixtures effects determination five different copper and glyphosate concentrations were preparead: 0,5μg/l Cu+ 0,5μg/l Gly; 2,5μg/l Cu +2,5 μg/l Gly; 5 μg/l Cu + 5 μg/l Gly; 50 μg/l Cu + 50 μg/l Gly; 500 μg/l Cu + 500 μg/l Gly. Terrestrial plants (Hordeum vulgare L.) were exposed to 5 different copper and glyphosate concentrations being 1000 times stronger than for aquatic plants: 0,01 mg /kg; 0,1 mg /kg; 1,0 mg /kg; 10,0 mg/kg; 100 mg/kg). At the same time the plants were cultured in a control solution (Steinberg medium) without the test substance. After 10 days of exposure Lemna minor and Hordeum vulgare in glyphosate and copper solutions, morphometric, physiological and biochemical duckweed and barley parameters were estimated. It was also estimated Lemna minor and Hordeum vulgare EC50 – concentration of each metal, causing 50% of the response to the effect and the concentrations of the mixture was expressed as toxic unit approach (TU). According to the method were estimated dry weight results to Lemna minor: Cu EC50 = 237,85 μg/l, Gly EC50 = 262,94 μg/l, Cu+Gly = 0,45 TU and Hordeum vulgare: Cu EC50 = 0,087 mg/kg, Gly EC50 = 5,41 mg/kg, Cu+Gly = 0,64 TU. It was investigated that copper was more toxic for aquatic plants (for dry mass), because lower concentrations leads a greater suppression of the dry mass compared with the same glyphosate concentration. The results of experiment showed that the greatest negative impact on the duckweed and barley growth, physiological and biochemical parameters had the strongest (1000μg /l and 100 mg/kg) concentration of the copper and glyphosate. Copper and glyphosate in a mixture stimulate each other: almost all the investigated cases were found synergistic interaction (TU<1). All tested concentrations had a significant impact on the oxidative stress on plants. Lemna minor and Hordeum vulgare had response already at the lowest concentrations of glyphosate and copper and accumulate larger amount of MDA than in the control group grew up plants. It was more sensitive aquatic plants.
