Abstract
The research was carried out at the Laboratory of Agrobio-technology of the Institute of Biology and Plant Biotechnol-ogy, Faculty of Agronomy of Vytautas Magnus University (till 2019 Aleksandras Stulginskis University). Theeffect of basal nutrition medium, subculture and growth regula-tors on the adventitious shoots formation from isolated ex-plants of Miscanthus× giganteus was analysed. Theapi-cal meristem of invitro developed adventitious shoots of Miscanthus× giganteus was cultivated in the Woody Plant Medium (WPM) and Murashige and Skoog (MS) nutrition medium supplemented with different 6-benzylaminopurine (BAP) and indole-3-butyric acid (IBA) or indole-3-acetic acid (IAA) combinations. It has been determined that the forma-tion frequency of adventitious shoots on the WPM nutrition medium was from 5.0 to 18.5% (after the first subculture) and from 19.5 to 38.3% (after the second subculture) higher in comparison with the MS nutrition medium supplemented with the same combinations of growth regulators. It has been established that on the WPM nutrition medium the formation frequency of adventitious shoots after the second subculture was from 13.3 to 18.5% higher in comparison with the first subculture, while on the MS nutrition medium the effect of subculture was not significant. Themost intensive frequency of shoots formation (48.8%) and the highest shoot number per explant (24.1 units) have been obtained in the WPM medium supplemented with 3.0 mg l–1 BAP + 0.4 mg l–1 IBA + 0.01 mg l–1 IAA after the second subculture.
Tyrimai atlikti Vytauto Didžiojo universiteto (iki 2019m. Aleksandro Stulginskio universiteto) Agronomijos fakulteto Biologijos ir augalų bio-technologijos instituto Agrobiotechnologijos laboratorijoje. Tirtas bazinės maitinamosios terpės, subkultivavimo ir augimo reguliatorių poveikis miskanto (Miscanthus×giganteus) pridėtinių ūglių formavimuisi invitro kultūroje. Eksplantai–in vitro kultūroje užaugintų pridėtinių ūglių apikalinės meristemos, auginti Woody Plant Medium (WPM), ir Murashigeir Skoog (MS) maitinamosiose terpėse be augimo reguliatorių ir su skir-tingais (1,0–4,0 mg l–1) 6-benzylamino purino (BAP) ir 0,4 mg l–1 indolil-3-sviesto rūgšties (ISR) bei 0,01 mg l–1 indolil-3-acto rūgšties (IAR) deri-niais. Įvertinus bazinės maitinamosios terpės poveikį ūglių proliferacijai invitro nustatyta, kad pridėtinių ūglių formavimosi dažnis WPM terpėje buvo nuo 5,0% iki 18,5% (po pirmo subkultivavimo) ir nuo 19,5% iki 38,3% (po antro subkultivavimo) didesnis, palyginti su MS terpe, papildy-ta analogiškais augimo reguliatorių deriniais. Įvertinus subkultivavimo po-veikį nustatyta, kad WPM maitinamojoje terpėje po antro subkultivavimo pridėtinių ūglių formavimosi dažnis buvo nuo 13,3% iki 18,5% didesnis, palyginti su pirmu subkultivavimu, o MS maitinamojoje terpėje esminių skirtumų tarp subkultivavimų nenustatyta. Intensyviausias (48,8 %) pridė-tinių ūglių formavimo dažnis ir didžiausias (24,1 vnt.) ūglių kiekis iš eks-planto nustatytas po antro subkultivavimo WPM maitinamojoje terpėje, papildytoje 3,0 mg l–1 BAP + 0,4 mg l–1 ISR + 0,01 mg l–1 IAR.
