Abstract
Magistrantūros studijų baigiamajame darbe pateikiama 2019–2020 m. Vytauto Didžiojo universiteto Žemės ūkio akademijos Agronomijos fakulteto Biologijos ir augalų biotechnologijos institute ir Atviros prieigos žemės ir miškų jungtinio tyrimų centro Agrobiotechnologijos laboratorijoje tirto augimo reguliatoriaus poveikis skiauteručio (Lophospermum D. Don) morfogenezei in vitro kultūroje tyrimo rezultatai.
Darbo objektas: skiauteručių (Lophospermum D. Don) veislės: 'Duchess Catherine'; 'Queen Elizabeth' ir 'Queen Morta'.
Darbo metodai: Izoliuoti eksplantai auginti Murashige-Skoog (MS) (Murashige, Skoog, 1962) mitybos terpėje be augimo reguliatorių (kontrolė) ir su skirtingais tidiazurono (TDZ) kiekiais: 0,1 mg L-1; 0,5 mg L-1; 1,0 mg L-1; 1,5 mg L-1; 2,0 mg L-1; 5,0 mg L-1.
Vertintas kaliaus susidarymo dažnis (balais), morfogenezės susidarymas (%) ir augimo indeksas.
Darbo rezultatai: Tiriant skiauteručio morfogenezes savybes, nustatyta, kad morfogenezės dažniui, kaliaus kiekiui bei augimo indeksui įtakos darė augalo genotipas ir augimo reguliatoriaus kiekis mitybos terpėje. Atlikti tyrimai rodo, kad 'Duchess Catherine' ir 'Queen Elizabeth' veislės yra reiklesnės mitybos terpei, ir joje esančiam augimo reguliatoriaus kiekiui, nei 'Queen Morta' veislė. Optimali tirto augimo reguliatoriaus – citokinino tidiazurono koncentracija skiauteručių morfogenezės formavimosi dažniui ir kaliaus indukcijai: 'Duchess Catherine' 0,5–1,0 mg L-1.; 'Queen Elizabeth' 1,0–1,5 mg L-1; 'Queen Morta' 1,5–2,0 mg L-1.

This master’s thesis presents the results of the study of the effect of growth regulator on the morphogenesis of creeping gloxinia (Lophospermum D. Don) in vitro culture during 2019–2020. The study was carried out at the Institute of Biology and Plant Biotechnology, Faculty of Agronomy of Vytautas Magnus University and at the Laboratory of Agrobiotechnology of the Open access Joint Research Centre of Agriculture and Forestry.
Research object: creeping gloxinia (Lophospermum D. Don) varieties, 'Duchess Catherine', 'Queen Elizabeth', and 'Queen Morta'.
Methods: isolated explants were grown in Murashige-Skoog (MS) (Murashige, Skoog, 1962) medium without growth regulators (control) and with different amounts of thidiazuron (TDZ): 0,1 mg L-1; 0,5 mg L-1; 1,0 mg L-1; 1,5 mg L-1; 2,0 mg L-1; 5,0 mg L-1. The frequency of callus formation (scores), morphogenesis formation (%) and growth index were evaluated.
Results: the analysis of the morphogenesis properties of creeping gloxinia showed that the amount of callus, the frequency of morphogenesis and the growth index were influenced by the genotype of the plant as well as amount of growth regulator in the culture medium. The study showed that varieties 'Duchess Catherine” and 'Queen Elizabeth' were more demanding on the nutrient medium and the amount of growth regulator it contains than the 'Queen Morta' variety. The optimal concentration of the analyzed growth regulator, cytokinin thidiazuron, for the induction of callus and frequency of morphogenesis formation: 'Duchess Catherine' 0,5–1,0 mg L-1.; 'Queen Elizabeth' 1,0–1,5 mg L-1; 'Queen Morta' 1,5–2,0 mg L-1.
